摘要
生物分子的识别、检测技术是生物分子学和生物医学研究中极为重要的领域。光波导模式谱(opticalwaveguidelightmodespectroscopy,OWLS)是一种非标记光学检测方法。OWLS方法通过检测波导附近的有效折射率的变化进行传感,无需进行放射性、荧光、或其他标记方法。通过同时对TM/TE两种传导模式的测定,可以实时定量分析波导表面吸附物质的质量。OWLS目前已经被应用到生物大分子相互作用、生物材料表征、环境及食品安全监测等领域。总结了以OWLS为原理构建的光栅生物传感器(opticalgratingcouplerbiosensor,OGCB)的原理,与其他非标记方法在传感指标方面的比较及应用领域的最新国外研究进展,并对该传感器的发展方向进行预测。
The detection of biomolecules is a very important part of biomedical research. Nowadays it is more popular to use a labeling method, e.g, isotope, fluorescence, etc. In the recent years, a non-labeling method called OWLS (Optical Waveguide Lightmode Spectroscopy), was rapidly developed. It has the features of non-labeling, and extremely high sensitivity, the convenience of manipulation and real-time in-situ measurement, which has the tendency to replace the labeling methods. In this paper, OWLS biochemical sensor was introduced, from its principle, comparison to labeling and other non-labeling methods, to its applications, development in recent years, and its evolution in the future is predicted.
