中药天年饮对衰老大鼠睾丸生精细胞增殖及相关因素的作用 被引量:9

Effects of Traditional Chinese Medicine Tiannianyin on PCNA Expression and Related Factors of Spermatogenic Cells in Testis of Aging Rats

摘要 目的研究中药天年饮对衰老大鼠睾丸生精细胞增殖及相关因素的影响。方法40只SD大鼠随机分为4组,正常组、衰老模型组、天年饮用药组及阴性对照组,每组均为10只。D-半乳糖连续腹腔注射制作亚急性衰老的大鼠模型。采用ELISA方法检测各组大鼠血清睾酮含量、SP免疫组化法检测睾丸生精细胞PCNA、端粒酶的表达情况。结果衰老大鼠血清睾酮含量、睾丸PCNA阳性细胞率及生精细胞端粒酶的表达明显低于正常大鼠(P<0.05,P<0.01)。天年饮可明显提高衰老大鼠血清睾酮含量(用药组与模型组比较P<0.05)、提高睾丸PCNA阳性细胞率(用药组与模型组比较P<0.01)及提高衰老大鼠端粒酶的表达。结论天年饮可明显改善衰老大鼠的生精功能,具有一定延缓衰老的作用。 Objective To observe the effects of traditional Chinese medicine Tiannianyin on PCNA expression and related factors of spermatogenic cells in testis of aging rats. Methods 40 SD rats were applied in this study and divided into 4 groups at random : normal group, aging model group, Tiannianyin group, negative control group, and each group had 10 rats. The sub-acute aging model rats were made by injecting D-gal into abdominal cavity continuousl. ELISA was used to detect the testosterone content in serum, SP immunohistochemistry to observe the PCNA and telomerase expression of spermatogenic cells in testis. Results The testosterone content in serum, the PCNA positive cell rate and the telomerase expression of aging rats were obviously lower than norreal rat (P 〈 0.05, P 〈 0.01 ). Tiannianyin could increase the testosterone content in serum ( P 〈 0.05 ), the PCNA positive cell rate (P 〈0.01 ) and the telomerase expression obviously. Conclusion: Traditional Chinese medicine Tiannianyin can improve the spermatogenic functions of aging rats obviously, and has fairly anti-aging effects.
