摘要
细菌的16 S核糖体RNA(ribosomeRNA,rRNA)以其在进化上的特征性序列,现已被广泛用于细菌分类和鉴定的分子指标。其具体操作是,以聚合酶链式反应(PCR)分离细菌样本中的16 S rRNA的基因片段,通过克隆、测序或酶切、探针杂交获得其序列信息,再与16 S rRNA数据库中的序列数据进行比较,确定其在进化中的位置,从而鉴定样本中可能存在的微生物种类。文章综述了16 S rRNA作为微生物系统分子分类鉴定的理论基础和方法,以及在细菌分类鉴定中的应用。
16 S rRNA gene is wildly used to classiy and identify of bacteria because of its special sequence in evolution. The detailed operation is that: the 16 S rRNA gene is isolated by means of polyrnerase chain reaction followed by sequence determination or digested by enzyme or hybrid. The sequence is compared with the date base to confirm the position of evolution. And then the bacterial sample is diagnosed. The paper reviewed the theory base of molecular identification by 16 S rRNA gene and application of bacterial classification.
