反相高效液相色谱法测定复方化学消毒剂中苯扎氯铵 被引量:32

Determination of Benzalkonium Chloride in Compound Chemical Disinfectants by Reversed-phase High Performance Liquid Chromatography

摘要 苯扎氯铵(BAC)主要由3种不同杀菌特性、正烷基(n—C12H25,n-C14H29和,n—C16H33)取代的二甲基苄基氯化铵组成。本文建立了反相高效液相色谱法测定复方化学消毒剂中苯扎氯铵的新方法。以乙腈-70mmol/L乙酸铵(含1%三乙胺,冰乙酸调PH至5.0)(体积比72:28)为流动相,Shiseido C18为分离柱,柱温30cc,262nm处检测,12rain内测定了样品中BAC的总量及n-C12H25-C9H13NCl、n—C14H29-C9H13NCl和n—C16H33-C9H13NCl各组分含量。n-C12H25-C9H13NCI、n-C14H29-C9H13NCl和n—C16H33-C9H13NCI的检出限分别为7.0、10、15mg/L(S/N=3),精密度(RSD)分别为0.6%、0.6%、1.1%,线性范围分别为0.04~1.6g/I.、0.04~1.6g/L和0.025—1.0g/L.低、中、高3种浓度水平的加标回收率为91%~99%。方法简单,适于测定复方化学消毒剂中BAC的总量及各单组分含量。 Benzalkonium chloride ( BAC ) is a mixture of alkylbenzyldimethylammonium chloride homologs with n-C12H25, n-C14H29 and n-C16H33 alkyl groups. A new reversed-phase high performance liquid chromatographic method(RP- HPLC) was established for the determination of BAC in combinational chemical disinfectants, n-C12 H25 - C9H13 NCl, n-C14 H29 - C9H13 NC1 and n-C16 H33 - C9H13 NCl were separated from the matrix on a Shiseido C18( 150 ×4. 6 mm, 5 μm) column using a mobile phase of V (A) : V(B) = 72 : 28 ( A: acetonitrile, B : 70 mmol/L ammonium acetate consisting of 1% triethylamine, adjusted with acetic acid to pH 5.0) and detected at 262 nm at 30 ℃. Good linear ranges of n- C12H25- C9H13 NCl, n-C14H29 -C9H13 NCl and n-C16H33 -C9H13 NCl were obtained from 0. 04 to 1.6 g/L, 0. 04 to 1.6 g/L and 0. 025 to 1.0 g/L with relative standard deviations( n =7 ) of 0.6% , 0.6% and 1.1% , respectively. Recoveries of three components at three different levels were in the range from 91% to 99%. The limits of detection were 7.0, 10 and 15 mg/L(S/N = 3), respectively. The method is simple and suitable for the accurate assay of both the total content of BAC and the content of each homologue in combinational chemical disinfectants.
