摘要
新鲜猪精液经乙醇沉淀,丙酮清洗,得抑制素粗提物,再经SephadexG-100凝胶过滤,收集蛋白质洗脱峰。采用小鼠hCG卵巢增重抑制法(改良斯蒂尔曼—波莱生物测定法)对其进行生物活性测定,粗品1000μg剂量组对性成熟前雌性小鼠卵巢增重的抑制作用极显著(P<0.01),第二峰下降部分200μg组的抑制效果显著(P<0.02)。表明从猪精液中可提取到具有生物活性的抑制素制品。
Fresh semen was precipitated with ethanol and washed with acetone to obtain a crude extract of porcine semen inhibin(psIB), and then purified by Sephadex G-100 Gel filtration to collect protein elution surge.The bioassay with hCG treated immature female mice by Modified Steelman Pohley method showed that the crude extract(dose 1 000 μg group) and the latter half fraction of the second surge (dose 200 μg group) significantly decreased the ovarian weight of hCG treated animals (P<0.01 and P<0.02). It seems that the preparation extracted from porcine semen has a biological activity of inhibin.
