摘要
目的探讨Bak、Fas基因在骨关节炎软骨细胞及滑膜中的表达及意义。方法成年新西兰大白兔24只,根据文献采用石膏固定方法制作骨关节炎动物模型,用免疫组化、原位末端标记技术(TUNEL)等方法检测Bak、Fas在兔膝关节软骨细胞及滑膜中的表达和软骨细胞凋亡情况。结果在软骨细胞中Bak、Fas的表达实验组阳性率均高于对照组(P<0.05);实验组软骨细胞凋亡程度与Fas、Bak的表达均呈正相关(P<0.05)。结论Bak、Fas基因与骨关节炎发生发展有密切关系。
[Objective] To investigate the expression and significance of Bak, Fas in articular cartilage and synovium of osteoarthritic animal model. [Methods] The left knees of 24 rabbits were immobilized in full extension using plaster cast. Immunohistochemistry and TUNEL assay was used to detect the expression of Bak, Fas proteins in articular cartilage and synovium and the apoptosis of the articular chondrocytes and synovial cell of animal model. [Results] The positive expression rat of Bak and Fas protein in osteoarthritic chondrocytes were higher than that in normal synovium (P 〈0.05). There were positive correlation between degrees of the expression of Bak/Fas and chondrocytes apoptosis in osteoarthritic articular cartilage (P 〈0,05). [Conclusion] There was a consanguineous correlation between degrees of the expression of Bak, Fas and chondrocytes apoptosis in osteoarthritic articular cartilage. It demonstrated that the abnormal expression of Bak, Fas closely related to the biological behaveior of OA such as occurrence, progress, et al.
