基因芯片分析银杏叶提取物对小鼠肾缺血再灌注损伤的保护作用机制 被引量:6

摘要 目的:用基因芯片分析银杏叶制剂(Ginkgo bilobaextract,EGb)对肾脏缺血再灌注损伤小鼠的基因表达影响,探讨其保护作用机制。方法:昆明种小鼠30只,随机分为假手术对照组、单纯缺血再灌注组、缺血再灌注+银杏叶提取物处理组。以香港城市大学基因组科技应用研究中心制作的包含7500个小鼠已知基因和ESTs的cDNA基因表达谱芯片,研究肾脏缺血45min再灌注24h的小鼠肾脏组织基因表达谱,并观察EGb对小鼠肾脏缺血再灌注损伤的基因表达影响。结果:单纯缺血再灌注组的小鼠肾脏组织基因共筛选出差异表达基因255条,其中115条基因表达上调,140条表达下调;缺血再灌注+银杏叶提取物处理组小鼠肾脏组织共筛选出差异表达基因244条,其中108条基因表达上调,136条表达下调。结论:银杏叶制剂对小鼠肾脏缺血再灌注损伤后的许多基因表达变化都有调节作用,这一综合性作用可能是其保护肾脏缺血再灌注损伤的机制。 OBJECTIVE To investigate the molecular mechanism of Ginkgo biloba extract (EGb) on prevention of acute ischemia-reperfusion injury in mouse by using eDNA microarray analysis. METHODS A model of renal ischemia-reperfusion injury was set up. 30 mice were randomly divided into sham operated group, isehemia-reperfusion group, and isehemia-reperfusion + EGb injection group. The renal gene expression profiles were tested using the gene chip from the City University of Hong Kong. And the effects of EGb on ischemia-reperfusion injury were analyzed. RESULTS Among the total 7 500 tested genes, the expressions of 115 genes were up-regulated and 140 genes were down regulated in ischemia-reperfusion group when compared with sham operated group. It was found that 108 genes were up-regulated and 136 genes were down-regulated in ischemia-reperfusion group as compared with sham operated group. CONCLUSION The results indicated that regulating the genes expression may be the mechanisms of EGb on prevention of renal ischemia-reperfusion injury in renal ischemia-reperfusion mouse.
