重组腺相关病毒介导的CD151基因转移对大鼠心肌梗死后血流动力学的影响 被引量:3

摘要 目的通过将携带人CD151基因的重组腺相关病毒(rAAV)栽体注入缺血心肌,观察CD151对大鼠心肌梗死后血流动力学的影响。方法制作大鼠急性心肌梗死模型,将rAAV-CD151注入缺血心肌。4周后取注射部位心肌,western blot检测CD151的表达,用心导管测定心脏功能,评价外源CD151对心肌梗死后血流动力学的影响。结果术后4周,CD151组心肌组织CD151蛋白表达水平高于假手术组、时照组、GFP组(P<0.05);CD151组LVEDP与心肌梗死组、GFP组比较明显降低,LVESP及±dp/dtmax明显增加,CD151基因的导入能够改善心室舒张和收缩功能。结论rAAV-CD151可以有效地转染大鼠心肌组织,改善左室功能。 Objective To investigate the therapeutic potential of recombinant adeno - associated virus mediated CD151 (rAAV - CD151) gene for ischemic heart diseases and the effect of CD151 on hemodynamics. Methods Rat myocardial infarction model was formed by ligation of the anterior descending coronary artery, and rAAV - CD151 was injected into the ischemic myoeardiurn. At 4 wecks after coronary artery ligation, western blot analysis for CD151 were performed, Henlodynamic variables were monitored. Results Compared with the rAAV - GFP group and eontroll group,CD151 gene delivery significantly increased the expression of CD151 and significantly improved the hemodynamic variables, including increases in LVESP, ± dp/dtmax, and an decrease in LVESP. Conclusion This study suggested that CD151 could improve hemodynamics after myocardial infarction in rats.
