丹参酮ⅡA对EVC304细胞血管舒缩因子的影响 被引量:20

摘要 [目的]探讨丹参酮ⅡA对血管内皮细胞分泌的血管舒缩因子的影响,以阐明其抗动脉粥样硬化作用及机制。[方法]通过建立肿瘤坏死因子-α(TNF-α)诱导的人脐静脉内皮细胞株(EVC-304)细胞损伤模型,以抗氧化剂吡咯烷二硫代氨基甲酸盐(PDTC)做为西药对照,采用放射免疫方法及硝酸还原酶法检测丹参酮ⅡA对EVC-304细胞培养液中血管舒缩因子的含量。[结果]丹参酮ⅡA能升高EVC304细胞培养液中舒血管因子一氧化氮(NO)的含量(P<0.05),降低缩血管因子内皮素(ET-1)和血栓素A2(TXA2)的含量(P<0.05或P<0.01);而对舒血管因子中前列环素(PGI2)的含量无明显影响(P>0.05)。[结论]丹参酮ⅡA可通过降低缩血管因子ET-1、TXA2的含量,增加舒血管因子NO的含量发挥其抗动脉粥样硬化作用。 [Objective] To illustrate the role of Tanshinone Ⅱ A(Tan Ⅱ A) which are extracted from dan-shen root in atherosclerosis (AS) and its different targets by investigating the effect on the diastolic and systolic factors of Human umbilical vascular endothelial cell 304(EVC304). [Methods] By establishing the model of TNF-α-induced cultured EVC304 cell, the radioimmunoassay and nitric reductase methods were used to analyze the content of the diastolic and systolic factors in the culture medium of the EVC304, which was cultured in different denses of Tan Ⅱ A. [Results] It was showed that the content of the diastolic factor such as NO was significantly enhanced by of TanlIA and the content of the diastolic factors such as ET-1 and TXA2 were induced significantly (P〈0.05 or P〈0.01). But it was not effect on the content of the diastolic factor of PGI2. [Conclusion] Tan Ⅱ A has a significant effect on the diastolic and systolic factors to suppress the progression of AS.
