摘要
目的研究在分离血小板中介入适量一氧化氮后血小板活化被抑制情况。方法用高纯铜与稀硝酸反应,制备NO气体,溶解于蒸馏水中制备成NO饱和溶液。手工制备富含血小板血浆,在滤过白细胞前介入不同浓度饱和NO溶液,将血小板最大回收率时的NO溶液浓度定为实验的最适浓度,并观察此NO浓度介入下保存期内血小板CD62p再表达率变化。结果NO饱和溶液进行10^(-2)稀释时,血小板的回收率最大。保存1d后cD62p再表达率差异有显著性(P<0.01)。结论一氧化氮可以减轻血小板体外活化程度。
Objective To observe the inactivation effects of nitrous oxide (NO) on plateleta zeparated from whole blood. Methods Platelet - rich plasma (PRP) was separated into aliquots firstly, copper and diluted hydrochlorie acid were uzed to produce NO in the environment of argon air, different diluted NO solutions were then infused into PRPs before leukocyte was filtered, the optimal NO concentration was achieved when the highest platelet recovery appearod. In the ease of optimum NO concentration, CD62p labeled with anti - CD62p - FITC was analyzed by FCM. Significant differences in the mean values between treatment groups and control groups were analyzed by paired t- teat for dependent sasaples. Results Highest platelet recovery was achieved when the dilution of NO saturate solution was 1/100, Significant difference was obaerved in the rate of CD62p re - expression one day after storage ( P 〈 0.01 ). Conclusion Infusion of authent/e NO into plateleta is an diicacioua method on the inhibition of platelets vitro activation.
