食用白酒灌胃后大鼠胃黏膜的抗损伤作用 被引量:3

Adaptive cytoprotection of gastric mucosa in rats after intragastric administration of edible alcohol

摘要 目的:观察食用白酒灌胃后,大鼠胃黏膜对无水乙醇损伤的反应及前列腺素E2(PGE2)、转化生长因子α(TGF-α)的表达情况.方法:SD大鼠36只,分别采用食用白酒530±20mL/L(A组),乙醇530mL/L(B组)、生理盐水(C组)按5mL/kg隔日给大鼠灌胃,在1wk末、1mo末处死动物,对比观察胃黏膜的Guth积分;用免疫组织化学方法测定胃黏膜中TGF-α的表达强度.另取36只大鼠,按以上相同处理后用酶联免疫方法测定胃黏膜中PGE2水平.结果:胃黏膜损伤程度由轻到重依次为A,B,C组,A,B,C组1wkGuth积分分别为26.5±6.4,40.5±8.1,69.3±4.3,两两比较均有统计学差异(P<0.05);A,B,C组1moGuth积分分别为34.8±8.0,54.8±8.0,74.8±3.1,两两比较均有统计学差异(P<0.05).1wkA,B组胃黏膜组织内PGE2水平与C组相比明显增高(17.2±1.8,16.6±1.1ng/Lvs13.0±1.5ng/L,P<0.05),A组与B组相比无差异(P>0.05);1mo后A组PGE2值下降,与C组相比无差异(P>0.05),而A,C组与B组相比均有差异(15.7±1.4,14.6±1.9ng/Lvs18.5±2.7ng/L,P<0.05).TGF-α表达1wkA,B组与C组有统计学差异(均P=0),A组与B组无差异;1moA,B,C组两两比较有差异(P=0.013,0.001,0).PGE2水平、TGF-α表达强度1wk与1mo比较,在A,C组差异有统计学意义(P<0.05);B组前后无差异.结论:食用白酒及乙醇对胃黏膜均可产生抗损伤作用;同时胃黏膜PGE2水平增高、TGF-α表达增强,PGE2,TGF-α在胃黏膜适应性保护过程中起到重要作用. AIM: To observe the response to edible alcoholinduced injury and expression of prostaglandin E2 (PGE2), transforming growth factor-α (TGF-α) in gastric mucosa of rats. METHODS: A total of 36 Sprague Dawley rats were divided group A, B, and C, intragastrically treated with edible alcohol (530 ± 20 mL/L), ethanol (530 mL/L), and normal saline at 5 mL/kg once every other day, respectively. The rats were killed at 1 week and 1 month after treatment, and the extent of mucosal injury was evaluated. Immunohistochemistry was used to examine the mucosal expression of TGF-α protein. Anther 36 rats were selected and treated as the method mentioned above, and the level of PGE2 in gastric mucosa was determined by enzyme linked immunosorbent assay. RESULTS: The most severe gastric mucosal injury was found in group C, then in group B, and the rats in group A were found with the mildest mucosal injury. The scores for mucosal injury were significantly different among group A, B and C 1 week (26.5 ± 6.4 vs 40.5 ± 8.1 vs 69.3 ± 4.3, all P 〈 0.05) and 1 month (34.8 ± 8.0 vs 54.8 ± 8.0 vs 74.8 ± 3.1, P 〈 0.05) after treatment. The level of mucosal PGE2 was increased significantly in group A and B as compared with that in group C 1 wk after treatment (15.7 ± 1.4, 14.6 ± 1.9 ng/ L vs 18.5 ± 2.7 ng/L, P 〈 0.05), but there was no difference between group A and B. One month after treatment, the level of PGE2 was decreased in group A, not different from that in group C, but there was obvious difference between the former two groups and group B (15.7 ± 1.4, 14.6 ± 1.9 ng/L vs 18.5 ± 2.7 ng/L, P 〈 0.05). The expression of TGF-α was significantly lower in group A and B than that in group C 1 week after treatment (both P = 0), but no difference was found between group A and B. TGF-α expression was significantly different among group A, B, and C 1 month after treatment (P = 0.013, 0.001, and 0, respectively). The level of PGE2 and the expression of TGF-α were notably different between 1 week and 1 month in group A and C, but not in group B. CONCLUSION: Appropriate stimulus with edible alcohol and ethanol may lead to adaptive cytoprotection in gastric mucosa through up- regulation of PGE2 and TGF-α.
