摘要
目的:探讨特发性血小板减少性紫癜(ITP)的B淋巴细胞的细胞周期分布和ITP的关系。方法:用碘化丙啶DNA染色法染色后用流式细胞仪检测40例初发的ITP患者治疗前、治疗恢复正常后及20例正常人的B淋巴细胞的细胞周期的分布比例,比较初发ITP患者治疗前、治疗恢复正常后与正常人的细胞周期分布的差异。结果:初发ITP组G0/G1期B淋巴细胞比例是(81.70±5.53)%,较治疗后恢复正常(90.38±0.92)%和对照组(91.53±2.09)少,差异有显著性(P<0.05);初发ITP组S期B淋巴细胞比例是(10.23±4.11)%,较治疗后恢复正常(5.60±0.91)%和对照组(5.18±1.21)%高,差异有显著性(P<0.05);初发ITP组G2/M期B淋巴细胞比例是(8.33±3.43)%,较治疗后恢复正常(4.03±1.76)%和对照组(3.30±1.84)%高,差异有显著性(P<0.05)。治疗后恢复正常和正常组比较G0/G1期、S期、G2/M期B淋巴细胞无差异(P>0.05)。结论:初发ITP患者治疗前S期和G2/M期B淋巴细胞比例增加,提示ITP患者的B淋巴细胞有过度增殖的现象,抑制ITP患者B淋巴细胞的细胞周期的转换可能是治疗ITP的新的治疗途径。
Objective: To explore the relationship between idiopathic thrombocytopenic purpura (ITP) and cell cycle distribution of B lymphocytes. Methods: DNA of B lymphocytes extracted from 40 patients with ITP before and after treatment and from 20 normal subjects was stained with propidium iodide, and the cell cycle distribution was analyzed by flow cytometry. Results: The percentage of B lymphocytes in G0/G1 phase (81.7±5.53) % was significantly lower than in the control group (91.53±2.09) % and ITP group after treatment(90.38±0.92) % ;the percentages of B lymphocytes in the S and G2/M phase (10.23±4.11 and 8. 33±3.43, respectively) were significantly higher than those in the control group (5.18±1.21 and 3.3±1.84, respectively) and ITP group after treatment (5.6±0.91 and 4.03±1.76, respectively). Conclusion: The proportions of S and G2/M phase of B lymphocytes before treatment were increased,which indicated that the B lymphocytes of the patients attacked by ITP had over-proliferated. It may be a new method to inhibit the transition of B lymphocyte cell cycle in ITP patients.
