硫磷铁试剂光度法测定鸡蛋中胆固醇含量的探索 被引量:20

摘要 本文用无水乙醇提取鸡蛋中的胆固醇,同时沉淀蛋白质,向提取液中加入高铁—磷酸—硫酸(硫磷铁)显色剂,胆固醇与浓硫酸及三价铁作用,生成较稳定的紫色化合物。测定波长为550nm,胆固醇含量在0~80μg/4ml范围内服从比尔定律,相对标准偏差为0.82%,回收率在98%~101%之间。本方法简便易行,测定鸡蛋中胆固醇的含量能获得满意的结果。 In this experiment,the cholesterol in eggs is extracted by alcohol with precipitating albumin,and then ferric chlorine-phosphoril and sulphuric acids (S-P-Fe) reagents are added to the extracted alcohol.The reaction of cholesterol with H2SO4-Fe3+ produces a more stable compound.It is shown that the reaction coincides Beer' s law under the wavelength of 550nm and the concentration of cholesterol from 0 to 80ug/4ml.The relative standard deviation of the results is 0.82%,and the recovery ratio of cholesterol is 98%~101%.So it is indicated that this method is simple and available for determining the cholesterol in eggs with satisfactory results.
