摘要
目的:探讨参附注射液预处理对心肌缺血再灌注损伤的保护作用及机制。方法:30只Wistar大鼠随机分为3组:假手术组、缺血再灌注损伤(IR)组、参附注射液预处理组(SFI)。建立大鼠心肌缺血再灌注损伤模型,应用原位杂交技术和免疫组化S-P法检测HSP70mRNA及其蛋白的表达。结果:与缺血再灌注损伤(IR)组比较,参附注射液预处理组(SFI)HSP70mRNA(P<0.01)及其蛋白(P<0.05)的表达显著增加。结论:HSP70是心肌中重要的内源性保护物质,参附注射液可诱导HSP70mRNA及其蛋白的表达而发挥心肌保护作用。
Objective: To investigate the protective effects and its mechanism of shenfu injection on myocardial ischemia reperfusion injury rats. Method: 30 Wistar rats were divided into three groups randomly: sham operation group, ischernia reperfusion injury group (IR), and shenfu injection preconditioning group (SFI). The model of myocardial ischemia reperfusion injury was produced. In situ hybridization assay and immunohistochcmical S- P method were used to detect the expression of HSP70 mRNA and its protein in 30 cases ischemia reperfusion injury myocardial tissues. Result: The expressions of HSP70 mRNA ( P 〈 0.01) and its protein ( P 〈 0.05) were significantly higher in shenfu injection preconditioning group (SFI) than that in ischemia reperfusion injury group (IR). Conclusion: HSP70 are important substabces of endogenous peotection, and shenfu injection can enehance the expressions of HSP70 gene and protect myocardial ischemia reperfusion injury.
