摘要
目的探讨荷人胃癌皮下移植瘤及细胞免疫功能重建的裸鼠模型构建方法及其特征。方法10只裸鼠随机分为2组,实验组(n=5)腹腔注射人外周血T淋巴细胞(HuPBTL)并皮下接种人胃癌细胞,对照组(n=5)仅皮下接种人胃癌细胞。观察肿瘤潜伏期、成瘤率和肿瘤体积,流式细胞仪检测裸鼠外周血中人T细胞,MTT法检测实验组鼠脾T细胞体外杀伤作用。皮下瘤行病理组织学检查。结果两组肿瘤潜伏期无统计学差异(P>0.05),成瘤率均为100%。第3周末,两组肿瘤体积相比无统计学差异(P>0.05);实验组鼠外周血中人CD3+、CD4+和CD8+T细胞分别为46.86%±9.14%、31.68%±10.17%和17.91%±1.38%,而对照组鼠外周血中则无人T细胞;当效靶比为20∶1和40∶1时,实验组鼠脾T细胞的特异性杀伤率分别为15.37%±1.58%和19.86%±2.86%,且随着效靶比增高而增强(P<0.05)。两组荷瘤鼠均无肝、肺转移;胃癌细胞保持亲代细胞的形态学特征。结论采用腹腔注射HuPBTL和皮下接种人胃癌细胞的方法成功构建了人胃癌-HuPBTL-裸鼠模型,能模拟具有一定细胞免疫功能的胃癌患者的荷瘤状态。
Objective To reproduce and characterize the a nude mice model with xenografts of human gastric carcinoma with reoonstruction of function of cellular immunity. Methods Ten nude mice were randomly divided into experiment group and control group (5 for each). In experiment group, mice were inoculated subcutaneously (s. c. ) with human gastric carcinoma cell BGC-823 and injected intraperitoneally (i. p. ) with human peripheral blood T-lymphocytes (HuPBTL). In control group, mice were inoculated s.c. with BGC-823 only. The tumor incidence, latency period, tumor volume were then monitored daily. The ratio of human CD3^+ , CD4^+ and CD8^+ T cell were detected by flow cytometry. Cytotoxic T-lymphocyte activity against autologous tumor cells was measured by MTT method. The pathohistological examination of tumor tissue of each mouse was performed. Results The latent period was 11.2±1. 9 days and 9. 4 ±1.5 days, respectively, for two groups. There was no significant difference between the two groups (P〉0. 05). The tumor incidence was 100% for both groups. At the third week after inoculation, tumor volume was 1 495. 6±111. 6mm^3 and 1 649. 6±128. 0mm^3 , respectively, and there was no significant difference between the two groups (P〉0. 05). The ratio of human CD3^+ , CD4^+ and CD8^+ T cells were 46. 86%± 9. 14%, 31.68%±10. 17% and 17. 91%±1.38% in experiment group, whereas human T cells could not found in control group. In experiment group, specific cytotoxicity were 15. 37%±1.58% and 19. 86%±2. 86% (P〈0. 05), E/T ratio was 20/1 and 40/1, respectively. Both groups showed No liver or pulmonary metastasis could be found in both groups. The pathohistological examination showed that gastric carcinoma cell preserved morphological characteristics of parental turental cells. Conclusion A novel nude mouse model with xenografts of human gastric carcinoma-HuPBTL was successfully reproduced by s.c. inoculation of human gastric carcinoma cells and i.p. injection of HuPBTL. It is a well-established animal model with transplantation of human gastric carcinoma that mimic clinical patients carrying the tufter.
