摘要
以小鼠脾脏T淋巴细胞的增殖活性为指标,考察中药芪苓制剂血清药理学试验中的给药剂量、给药天数、血清添加剂量和孵育时间等最佳条件.结果表明:按小鼠每公斤体重给生药0、12.5、25.0、50.0 g连续3和6 d,25.0 g.kg-1的剂量对小鼠脾淋巴细胞增殖的促进作用显著或极显著地优于0、12.5、50.0 g.kg-1(P<0.01或P<0.05),其中连续6 d给药的效果更好;体外细胞培养体系中加入体积分数为20%或40%的含药血清,与加入5%、10%、80%、100%含药血清比较,能够极显著促进小鼠脾淋巴细胞的增殖(P<0.01),其中以添加20%的效果更好,孵育2 d比孵育3 d的效果更好.
The proliferation activity of spleen T lymphocytes of mice was taken as index in the study which aimed at exploring the best condition of medicine dosage, administration days, serum dosage and incubation time in the serum pharmacology test of compound radix astragali preparation (CRAP). The result indicated that using 25.0 g·kg^-1 CRAP of body weight to mice for 3 days and 6 days, the corresponding serum could improve the proliferation of spleen lympheeytes with significant differences (P 〈0.01 or P〈0.05), comparing with the 0, 12.5, 50.0 g ·kg^-1 dosages. The 6 days administration had better effect than 3 days administration. Adding 20% or 40% medicine-containing serum to the cell culture system could significantly improve the proliferation of spleen lympheeytes versus 5%, 10%, 80%, 100% serum adding amount, and 20% serum had better effect. 2 days incubation had better effect than 3 days incubation.
