芪玄益心胶囊对糖尿病大鼠急性缺血心肌NF-κBp50、IκBα、IL-1β及其mRNA的影响 被引量:5

Influence of Qixuanyixin Capsules on expression of NF-κBp50,IκBα and IL-1β protein and mRNA of acute ischemic cardiac muscle in diabetic rats

摘要 目的探讨芪玄益心胶囊对糖尿病(DM)大鼠急性缺血心肌保护作用的机制。方法以四氧嘧啶腹腔注射致胰腺损伤,舌下静脉注射垂体后叶素致心肌急性缺血复制大鼠DM急性心肌缺血模型;将其随机分为美吡达组、丹参滴丸组、芪玄益心胶囊高中低剂量组、模型组,并设空白对照组。观察各组模型心肌核转录因子-κBp50(NF-κBp50)及其抑制蛋白(IκBα)、白介素-1β(IL-1β)及其mRNA水平的表达并与空白组对照。结果模型组及各治疗组与空白组比较NF-κBp50(细胞核)、IκBα(细胞浆)、IL-1β及其mRNA表达均显著增高(P<0.05)。芪玄益心胶囊各剂量组与模型组比较,NF-κBp50、IκBα、IL-1β及其mRNA表达明显降低(P<0.05)。芪玄益心胶囊与美吡达、丹参滴丸比较,对心肌NF-κBp50、IκBα、IL-1β及其mRNA改善优于美吡达与丹参滴丸(P<0.05)。结论糖尿病大鼠缺血心肌中存在NF-κBp50、IL-1β的激活,芪玄益心胶囊对其有一定的抑制作用,推测抑制NF-κBp50(IκBα)—IL-1β这一途径的激活,是芪玄益心胶囊心肌保护作用的重要机制之一。 objective To investigate the mechanism of protective effect of Qixuanyixin Capsules on diabetic rats with myocardial ischemia. Methods The diabetic rat models with myocardial ischemia were reproduced by injecting alloxan intraperitoneally to cause pancreas damage and injecting pituitrin in raninevein to produce myocardial ischemia, which were randomly divided into Minidiab group, Danshen Gutta Pills group, high dose of Qixuanyixin Capsules group, medium dose of Qixuanyixin Capsules group, low dose of Qixuanyixin Capsules group and model group. There was a blank control group with the same number of healthy Wistar rats. The expression of NF-κBp50, IκBα and IL-1β portein and mRNA of acute ischemic cardiac muscle in rats of all groups were detected. Results Compared with the blank control group, the expression of NF-κBp50, IκBαand IL-1β portein and mRNA of acute ischemic cardiac muscle were higher in the other groups( P 〈0.05 ). In high, middle and low dose of Qixuanyixin Capsules groups, the levels of NF-κBp50, IκBαand IL-1β protein and mRNA were lower than in the model group (P 〈 0.05 ), and the effect of Qixuanyixin Capsules was better than that of Danshen Gutta Pills and Minidiab(P 〈 0. 05 ). Conclusions The activation of NF-κBp50 and IL-1β lies in ischemic cardiac muscle of diabetic rats, which can be inhibited by Qixuanyixin Capsules to some degree. It is one of the important protective mechanisms of Qixuanyixin Capsules on cardiac muscle.
