α-黑色素细胞刺激素对ECV304细胞产生促炎因子的调节作用 被引量:1

Effect of α-melanocyte stimulating hormone on the regulation of the production of pro-inflammatory cytokines from human umbilical vein endothelia cell line

摘要 研究α-黑色素细胞刺激素(α-MSH)对人脐静脉内皮细胞系ECV304细胞产生促炎因子的调节作用,探讨α-MSH的抗炎与免疫调节机制。用不同浓度α-MSH处理ECV304细胞,或在LPS/TNF-α处理的同时加不同浓度的α-MSH培养ECV304细胞。用ELISA检测上清中TNF-α和IL-8,以Greiss法测定NO,RT-PCR检测组织因子(TF)mRNA,Westernblot检测TF蛋白表达。结果显示,α-MSH抑制ECV304细胞产生NO、TNF-α,但促进IL-8的释放。LPS或TNF-α能上调ECV304细胞表达TF mRNA,而α-MSH抑制这种上调作用,并抑制ECV304细胞表达TF蛋白。上述结果提示α-MSH可通过调节血管内皮细胞表达促炎因子而发挥抗炎和免疫调节作用。 The regulatory effect of α- melanocyte stimulating hormone(α-MSH) on the production of pro-imflammatory cytokines from human umbilical vein endothelial cell line (ECV304) was investigated to explore the anti-inflammatory and immunoregulatory mechanism related to α-MSH, in which ECV304 cells were treated with different concentrations of α-MSH,or with addition of LPS or TNF-α at the same time. The level of TNF-αor IL-8 in the supernatants of cell cultures was determined by ELISA and the concentration of nitric oxide(NO) was detected by Greiss method. Meanwhile, the tissue factor mRNA was tested by using RT-PCR and the tissue factor proteins were examined by Western blot assay. The experimental results showed that α-MSH could inhibit the production of NO and TNF-α,but promote the release of IL-8. In addition, LPS or TNF-α up-regulated the expression of tissue factor mRNA, and α-MSH could inhibit this process of up-regulation,and also the expression of tissue factor protins in ECV304 cells. These results indicate that α-MSH may exert anti-inflammatory and immuno-regulatory effects through regulating the expression the production of proinflammatory cytokines from vascular endothelial cells.
