摘要
根据大米草Na+/H+泵基因已克隆的一段基因组DNA序列的保守区设计引物,利用RACE技术,从大米草中分离并克隆出Na+/H+泵基因的一段cDNA片段,长度为793 bp.该cDNA片段编码区推导出的氨基酸序列在NCB I网站上BLAST,结果显示该氨基酸序列与多种植物的Na+/H+泵的C端氨基酸序列相似.该结果可以推断所克隆的cDNA片段为大米草Na+/H+泵基因的3′端.将该cDNA对应的氨基酸序列与以前克隆的大米草Na+/H+泵基因的一段DNA所对应的氨基酸序列拼接,然后与多种植物的Na+/H+泵进行对位排列.结果显示,该氨基酸序列与芦苇、水稻、小麦、大麦、玉米、拟南芥和滨藜等植物的Na+/H+泵基因对应的氨基酸序列有较高的相似性,其中与禾本科植物的相似性程度更高.
The study was to clone the 3′cDNA of Na^+/H^+ antiporter gene from English cordgrass by RACE using a series of primers designed according to a genomic DNA segment of English cordgrass cloned before. A segment of 793 bp in length was obtained. The sequence has high homologies to the Na^+/H^+ antiporter genes of many plants, suggesting that it might be really the 3′cDNA of the Na^+/H^+ anfiporter gone from English cordgrass. The amino acid sequence deduced from the cDNA was jointed with the amino acid sequence deduced from the gcnome DNA sequence cloned before. The new amino acid sequence was aligned with corresponding Na^+/H^+ antiporter amino acid sequence of some other plants. The result showed that the amino acid sequence is highly homologous to the Na^+/H^+ antiporter genes from flee, barley, wheat, cotton, Iris lacteal and other plants.
