摘要 目的观察自创增强酶解法提取的皮质骨胶原所制胶原膜的生物相容性,为利用该胶原制备支架材料提供可行性依据。方法以新鲜猪皮质骨为原料,通过增强酶解法提取技术获得Ⅰ型胶原,再将胶原制成戊二醛交联或不交联的胶原膜。最后将胶原膜与hMSC共培养观察细胞相容性或埋植到兔肌袋里观察组织相容性和降解情况。结果胶原膜无明显细胞毒性,hMSC增殖迅速,无明显受抑制现象。埋植试验,未交联的胶原膜1周时即明显降解,可见血管长入,2周末即被完全吸收;交联后的胶原膜2周时开始明显降解,4周左右才完全被吸收。结论增强酶解法提取的骨胶原制备的胶原膜具有良好的细胞相容性、组织相容性和降解性。适当浓度戊二醛交联可提高胶原的强度而不引起明显的毒性。 Objective To observe the biocompatibility of the bone-collagen membrane made with the collagen which was isolated by the enhanced pepsin digestion method, and offer the feasible evidence to prepare the scaffold. Methods Taking the cortical bone of porcine as material, Ⅰ-collagen was isolated by using enhanced pepsin digestion method. Last, observe the eelluar compatibility by culturing the hMSC with the collagen membranes together, and observe the histocompatibility and degradation after grafting the collagen membranes into the muscles of rabbit. Results Collagen membranes hadn't obvious cytotoxicity, and the hMSC hadn't been inhabited and proliferated quickly. In the graft test, the uncross-linked bone collagen membranes (CM) degradated obviously 1 week later, some blood tubes grew into them,and been absorbed completely after 2 weeks. While, the cross-linked bone collagen membranes (GLCM) by glutaraldehyde degradated obviously 2 weeks later,and been absorbed completely after 4 weeks. Conclusion The bone-collagen membranes made with the collagen which was isolated by the enhanced pepsin digestion method has a good celluar compatibility, histocompatibility and degradation ability. The collagen' s intensity can be improved by cross-linking with a proper concentration of glutaraldehyde, without obvious toxicity.
