摘要
目的检测雌性大鼠去卵巢后颏舌肌肌功能的变化,探索雌激素影响上气道稳定性的机制。方法30只雌性SD大鼠随机分成三组:正常组、假手术组、去卵巢组。手术6周后,麻醉动物,切断双侧迷走神经,采用插入式双极针电极引导大鼠颏舌肌肌电;电脉冲刺激舌下神经中央支诱发颏舌肌收缩。原始信号经SMUP-PC型生物信号处理系统输入计算机,使用MFlab功能学实验软件包Version3.00分析,检测各实验动物颏舌肌的肌电强度、最大等长收缩力和70%强直收缩力的时程。结果假手术组颏舌肌的肌电强度、最大收缩力、70%强直收缩时程与正常组相比无显著性差异(P>0.05),去卵巢组颏舌肌的肌电强度、最大收缩力、70%强直收缩时程较正常组明显降低(P<0.05)。结论雌性大鼠去卵巢后颏舌肌肌功能降低,雌激素可能通过增强颏舌肌肌功能的途径对上气道稳定性发挥保护作用。
Objective To examine the effects of ovariectomy on genioglossus (GG) muscle activity in female rats and to investigate the mechanisms underling hormone protecting the patency of upper airway (UA). Methods Thirty female Sprague- Dawley rats were randomly assigned to three groups: normal group( NORMAL), sham group (SHAM) and ovariectomized group (OVX). Six weeks later, the rats were anesthetized and vagotomied. Electromyography activity of the genioglossus (EMGgg) was obtained from inserted bipolar electrodes. The contractile characteristics of the whole GG were measured in response to stimulation of the hypoglossal medial nerve branch. Data were inputed SMUP - PC biomedical signal processing system and analyzed by MF lab functional experiment analysis software. The integrated EMG (iEMG), twitch tension and 75 percent - decay time of tetanic tension were analyed. Results EMGgg and the contractile properties of genioglossus had no significant variety between the normal and sham groups ( P〉 0.05 ). Compared to normal group, iEMG, twitch tension, 75 percent - decay time of tetanic tension significantly decreased in OVX group ( P 〈 0.05). Conclusion The genioglossus muscle activity in female rat decreased following ovarian hormone removal. Ovarian hormone may protect the patency of UA through affecting muscle function of upper airway dilator.
