摘要
目的：探讨低抗凝肝素对胎牛血清培养的大鼠肝星状细胞生长的影响。方法：肝星状细胞经体积分数为0.4%胎牛血清/DMEM培养液同步化48h后分为对照组和实验组,对照组以体积分数为10%胎牛血清处理,实验组分别以体积分数为10%胎牛血清和不同质量浓度（8g/L,40g/L,200g/L,1000g/L,5000g/L）低抗凝肝素处理,应用MTT法测定其对肝星状细胞生长的影响。结果：低抗凝肝素（40~5000g/L）对肝星状细胞的增殖有明显抑制作用（P〈0.05）。在不同的作用时间,低抗凝肝素（200g/L）均对肝星状细胞生长有明显抑制作用,且在其作用48h后肝星状细胞生长速度明显减慢。结论：低抗凝肝素对大鼠肝星状细胞生长具有抑制作用。
Aim ： To explore the effect of low anticoagulative activity heparin on the growth of rat hepatic stellate cells （HSCs）. Methods：Rat HSCs were treated with 0.4% fetal calf serum （FCS）/DMEM for 48 hours and divided into the control group and experiment groups. HSCs in the control group were treated with 10% FCS, and HSCs in the experiment groups were treated with 10% FCS and low anticoagulative activity heparin with different concentrations （ 8 g/L, 40 g/L, 200 g/L, 1 000 g/L, 5 000 g/L）. MTT reduction assay was used to evaluate the effect of low anticoagulative activity heparin on the growth of rat HSCs. Results： Low anticoagulative activity heparin with different concentrations （ 40 - 5 000 g/ L） all inhibited the growth of HSCs significantly. At different acting time points, low anticoagulative activity heparin （200 g/L） could inhibit the growth of HSCs significantly, especially after 48 hours. Conclusion： Low anticoagulative activity heparin can inhibit the growth of rat HSCs.
