摘要
目的:探讨新型砷剂配合物砷化砷-硫脲(AsI3-Tu)对白血病HL-60细胞增殖及survivin基因表达的影响。方法:以不同浓度的砷化砷-硫脲作用于HL-60细胞,应用噻唑蓝(MTT)比色法检测细胞增殖活性,原位末端标记(TUNEL)法观察细胞凋亡,RT-PCR半定量检测细胞中sur-vivin mRNA表达。结果:①各实验组AsI3-Tu均能抑制HL-60细胞增殖,并与时间和剂量相关。②TUNEL法观察细胞凋亡,可见AsI3-Tu各浓度组凋亡细胞数明显增多。③AsI3-Tu各浓度组细胞中survivin mRNA表达水平降低,且表达水平与AsI3-Tu剂量呈负相关。结论:AsI3-Tu能有效抑制白血病细胞系HL-60细胞的增殖,诱导其凋亡,机制可能与抑制HL-60细胞中survivin基因表达有关。
To investigate the effects of new arsenicals (AsI3-Tu) on proliferation and expression of survivin in HL-60 cells. Methods: HL-60 cells were treated with AsI3-Tu (0,1,2,4,8 μmol/ L) in vitro. Inhibitory effect of AsI3-Tu on proliferation of HL-60 cells was measured by MTT assay, and cell apoptosis was observed by TUNEL method. The expression of survivin mRNA was evaluated with semi-quantitatlve RT-PCR. Results : ①1~8μmol/ L AsI3-Tu could obviously inhibit the proliferation of HL-60 cells,and the survival rate of HL-60 cells was negatively correlated with time and dosage of AsI3-Tu. ②According to the result of TUNEL,it proved that the amount of apoptosis cells had been obviously increased in AsI3-Tu groups. ③The expression of survivin mRNA was down-regulated after treatment with AsI3-Tu and negatively correlated with the dosage of AsI3-Tu. Conclusion : AsI3-Tu can effectively inhibit the proliferation of HL-60 cells and induce apoptosis. The mechanism may be related with the expression of survivin down-regulated .
