海风藤抑制淀粉样前体蛋白表达的研究

Caulis Piperis Kadsruae negatively regulates the expression of amyloid precursor protein in SK-N-SH Cells

摘要 目的研究中药海风藤对人类神经母细胞瘤细胞系SK-N-SH淀粉样前体蛋白(APP)基因表达的影响。方法MTT比色法检测海风藤作用后SK-N-SH细胞活性,RT-PCR和Western blot方法分别检测APP基因mRNA及蛋白质的表达。结果海风藤能够选择性降低SK-N-SH细胞APP mRNA表达,且呈药物浓度依赖性,海风藤对APP几种同功异构体的表达均有不同程度的抑制作用。在实验药物浓度内,海风藤抑制APP的表达,未导致SK-N-SH细胞活性下降。结论海风藤能够选择性抑制SK-N-SH细胞系APP的表达。 Objective To investigate the effects of Canlis Piperis Kadsruae on amyloid precursor protein (APP) gene expression in human neuroblastoma cell line SK-N-SH. Methods Cell viability was tested by MTT assay, the expressions of APP mRNA and protein isoforms were quantified by RT-PCR and Western blot analysis, respectively. Results Caulis Piperis Kadsruae caused a significant decrease in the total APP mRNA band in a dose dependent manner, and the expressions of the intracellular immature and mature APP in SK-N-SH cells were inhibited by Caulis Piperis Kadsruae. In the experimental condition, Caulis Piperis Kadsruae (0 -50 g/L) did not reduce the survival rate of SK-N-SH cells. Conclusion Caulis Piperis Kadsruae can selectively inhibit the expression of APP in SK-N-SH cells.
