摘要
观察藤梨根正丁醇提取物对培养的人食管癌Eca-109细胞的生长抑制作用,探讨藤梨根对人食管癌细胞的作用机制。方法:MTT比色法检测藤莉根正丁醇提取物在不同浓度(1μg/ml、10μg/ml、100μg/ml)及不同时间(24 h、48 h、72 h)对Eca-109细胞生长抑制作用,TUNEL法检测其对癌细胞生长的诱导凋亡效应。结果:藤梨根正丁醇提取物对人食管癌Eca-109细胞的生长抑制作用随着药物浓度的升高和作用时间的延长而增强,其生长抑制率可达到87.2%。同时,提取物对瘤细胞有明显的凋亡效应,而在对照组,未见有明显凋亡现象。结论:藤梨根正丁醇提取物能有效抑制人食管癌Eca-109细胞生长。
Objective: To study the inhibitory effect of extracts from Actinidia arguta by n-butyl alcohol on human carcinoma of esophagus cells(Eca-109) and Its mechanism. Methods: MTT colonmetric assay was used to examine the growth inhibitory of concentration-effect( 1 μg/mL,10 μg/ml,100 μg/ml) and time-effect (24 h,48 h,72 h) of extracts from Actinidia arguta by n-butyl alcohol on Eta-109 cells. TUNEL test were performed to observe the apoptosis induced by the extracts ( 1 μg/ml, 10 μg/ml, 100 μg/ml) on Eca-109 cells. Results: The inhibitory effect of the extracts on Eca-109 cells increased in a dose-time Manner and the highest rate of inhibition was 87. 2%. The extracts could significantly induce apoptosis of Eta-109 cells, but in control group, apoptosis wasn't observed. Conclusion: The extracts from Actinidia arguta by n-butyl alcohol have good inhibitory effect on Eta-109 cells.
