摘要
目的研究Rac1和缺氧诱导因子1α(hypoxia inducible factor1α,HIF-1α)在激光诱导实验性小鼠脉络膜新生血管(choroidal neovascularization,CNV)模型组织中的表达及意义。方法用532nm激光诱导小鼠CNV模型,分别在光凝后1d、3d、7d、14d、28d行荧光素眼底血管造影后处死小鼠,摘除眼球制作标本,行病理切片观察及免疫组织化学检测Rac1和HIF-1α蛋白在CNV组织中的表达。结果正常小鼠视网膜脉络膜组织未见Rac1和HIF-1α蛋白阳性表达。激光诱导的小鼠CNV组织形成过程中均有Rac1和HIF-1α蛋白的表达,激光光凝早期二者表达均增高;光凝后1d即有明显的阳性表达,二者的阳性表达率分别是11.21%和24.80%;光凝后3~7d均达到高峰,Rac1蛋白的阳性表达率分别为34.58%和53.35%,而HIF-1α蛋白的阳性率为33.05%和50.35%;光凝后14d,二者的表达均下降,阳性率分别为25.50%和31.97%;光凝后28d,阳性率分别为9.32%和11.26%;二者表达呈显著正相关(r=0.943,P=0.016)。结论Rac1和HIF-1α在CNV组织中存在高表达,且二者表达密切相关。
Objective To study the expression of Racl and hypoxia induc- ible factor-1α( HIF-1α) in tissues of experimental choroidal neovaseularization (CNV) model in mice and its significance. Methods CNV models were induced by 532 nm laser,the eyeballs were taken out on the lst,3rd,7th,14th and 28th day respectively after fluorescein angiography was performed. The expression of Racl and HIF-1α in CNV tissues was detected by imnaunohistochemistry. Results Racl and HIF-1α proteins were negative in normal retinal and choroidal tissues,but which were both significantly expressed in tissues of experimental CNV model induced by 532 nm laser. Their expression both increased during the earlier period after photocoagulation. The positive expression rates of Racl and HIF-1α were 11.21% and 24.80% respectively on the 1st day after laser,and achieved the peak on the 3rd and 7th day,when the positive expression rates of Racl were 34.58% and 53.35% , and that of HIF-1α were 33.05% and 50.35% respectively. On the 14th day, the expression of Racl and HIF-1α both decreased,the rates of which were about 25.50% and 31.97% respectively. On the 28th day, the positive expression rates of Racl and HIF-1α were 9. 32% and 11.26% respectively. There was obvious correlation between the expression of Racl and HIF-1α(r=0.943,P=0.016).Conclusion Racl and HIF-1α are significantly expressed in tissues of experimental CNV model,and one is closed correlated with the other.
