摘要 目的:观察骨髓源性干细胞能否向慢性马兜铃酸肾病大鼠肾脏细胞分化及移植后相关生化指标的动态变化。方法:制备雌性Wistar大鼠CAAN动物模型,亚致死剂量X线照射后经尾静脉移植同系雄性大鼠骨髓干细胞,于移植后第60d处死大鼠,留取血、尿及肾组织标本。生化检测观察移植后相关生化指标的改善情况,Y染色体荧光原位杂交(Y-FISH)观察雌性受体大鼠肾脏中雄性骨髓源性干细胞的分化情况。结果:骨髓移植60d后在肾小球及肾间质中均发现Y染色体阳性的供体源性细胞,相关生化指标得到改善。结论:骨髓源性干细胞具有向慢性马兜铃酸肾病大鼠肾小球及间质细胞分化的潜能。 Objective: To investigate whether bone marrow-derived stem cells can differentiate into renal cells in rats with chronic aristolochic acid nephropathy (CAAN) and the dynamic changes in the biochemical indicators after the transplantation of bone marrowderived stem cells. Methods: CAAN model was established in female Wistar rats, The bone marrow-derived stem cells from homologous male Wistar rats were transplanted into CAAN rats via the caudal vein after sublethal dose irradiation of X-ray. CAAN rats were killed 60 days after the transplantation, and the specimens of blood, urine, and renal tissues were obtained. The changes in biochemical indicators were observed, and the differentiation of the bone marrow-derived stem cells was detected by using Y-chromosome fluorescence in situ hybridization (Y-FISH). Results:Sixty days after the transplantation, Y-chromosome-positive cell derived from donor rats was found in the glomerulus and renal interstitium of recipient rats, and the biochemical indicators were improved. Conclusion: The bone marrow-derived stem cells could differentiate into glomerular cells and renal interstitial cells in CAAN rats.
