摘要
目的探讨Synphilin-1对帕金森病(PD)酪氨酸羟化酶(TH)表达的影响。方法50只SD大鼠随机分成5组,每组10只。PD组大鼠右侧黑质注入6-羟多巴3μL制作PD模型,反义组、同义组、错义组分别注入Synphilin-1反义寡核苷酸、同义寡核苷酸、错义寡核苷酸各3μL,正常对照组右侧黑质注入2 g/L抗坏血酸生理盐水。取鼠的术侧脑黑质制作切片,利用免疫组化方法检测Synphilin-1表达,利用免疫组化、Western blot、原位杂交方法检测TH的表达。结果与正常对照组比较,PD组Synphilin-1蛋白表达显著增加,TH蛋白和TH mRNA表达显著减少;与PD组比较,Synphilin-1反义组TH蛋白和TH mRNA表达显著增多。结论Synphilin-1可能通过影响TH的表达对帕金森病的发病机制产生影响。
Objective To investigate the effect of synphilin-1 on tyrosine hydroxylase (TH) expression of Parkinson di sease. Methods Fifty healthy male Sprague-Dawley rats were divided evenly into five groups. PD group. PD models were made by unilateral stereotaxic infusion of 3 μL of 6-hydroxydopamine (6-OHDA) in right substantia nigra. Antisense group, sense group and missense group were given antisense, sense, and missense oligonucleotides respectively. Control group: natrium saline was injected into right substantia nigra. The expression of synphilin -1 protein was detected by immunohistochemistry, and expression of TH protein and mRNA immunohistochemistry, western blot, and in situ hybridization. Results Compared with control group group, the expression of synphilin- 1 in PD significantly increased, and the expression of TH decreased. Compared with PD group, the expression of TH protein and TH mRNA in antisense group significantly increased. Conclusion Synphilin -1 may affect the pathogenesis of PD by affecting the TH expression.
