心脏移植后急性排异反应体外模型的建立及其应用研究 被引量:2

STUDY ON SETING UP OF A MODEL OF ACUTE REJECTION OF HEART TRANSPLANTATION IN RATS IN VITRO

摘要 分离成年Wistar大鼠心肌细胞(2×106ml-1)SD大鼠的脾细胞(1×107ml-1),前者做刺激细胞,后者做反应细胞,进行混合细胞培养,建立大鼠心脏移植后急性排异反应体外模型。该模型包括4组:①阴性对照组,心肌细胞与经丝裂霉素C处理的脾细胞共同培养。②阳性对照组,心肌细胞与脾细胞共同培养。③药物对照组,在阳性组基础上加入CSA(环孢霉素A10μg/ml)。④实验组,在阳性组基础上加入实验药物(HSP,人类精浆,10μg/ml)。应用光镜、电镜观察各组心肌细胞的受损情况和脾细胞转化为淋巴母细胞的数量,并用3H-TdR掺入法测定脾细胞的转化率,证实了CSA和HSP有相似的免疫抑制作用。用大鼠腹腔内心脏异位移植体外模型来重复验证CSA和HSP的免疫抑制作用,两者结果相同,从而证明了心肌细胞混合培养的体外模型不仅简便易行,可靠性强,而且为大规模地筛选免疫抑制剂提供了一种方便、廉价的实验工具。 Cardiomyocytes of adult Wister rats and spleen cells of adult SD rats were separated,and the cardiomyocytes(2×10 6 ml -1 )as stimulate cells and spleen cells(1×10 7 ml -1 )as responsed cells were cultured in 96 culture plated.The model of acute rejection of cardiac transplantation of rats in vitro was set up.The model includes four groups,negative group(mixed culture of the cardiomyocytes and the spleen cells treated with mitomycin C),positive group(mixed culture of the cardiomyocytes and the spleen cells),drug control group(cyclosprione A(CSA)group,mixed culture of the cardiomyocytes,the spleen cells and CSA)and experiment group(mixed culture of the cardiomycytes and the spleen cells and HSP(human seminal plasma)).Observed with microscope and electromicroscop,the damages of cardiomyocytes of the all of groups,and blastgesis of the recipients spleen cells(responsed cells)induced by donors cardiomyocytes(stimulate cells)with 3H thymidine.The authors confirmed immunosuppression activity of CSA and HSP with the model in vitro and model in vivo of cardiac transplantation in rats.The results of in vitro and in vivo experments are identical.It is concluded that the model in vitro can immitate well acute rejection of cardiac allo graft in vivo.It is a good model in the study of the mechanism of acute rejection in heart transplantation.This model has advantage of convenience,dependence,etc.
