摘要
采用农杆菌介导的遗传转化法,将抗除草剂Bar基因转入籼稻品种明恢63,共获得9个独立转化子,其中3个为单拷贝。PCR和Southern blot表明,Bar基因已经整合到水稻基因组上。3个单拷贝转化子的纯合系通过发芽试验获得且均表现对Basta具有高度抗性。这些材料可直接生产抗除草剂杂交种,也可作为抗除草剂的种质资源,通过杂交、回交等常规育种方法将抗除草剂Bar基因导入目标品种,获得具有抗除草剂特性的新品种。
Herbicide-resistant Bar gene was introduced into Minghui 63 via Agrobacterium-mediated process and a total of 9 independently transformants was obtained, 3 of which had single-copy insertions. Integration of Bar gene into rice genome was conformed through PCR and Southern blot. Homozygous families was obtained by seed germination and all these homozygous families showed highly resistance against herbicide Basta. These single-copy transformants could be used for the culture of resistance-herbicide and also produce new variety through backcrossing and crossing.
