丝裂霉素联合卡介苗激活的淋巴细胞对膀胱肿瘤细胞株TBC-1杀伤作用的实验研究

摘要 ①目的 探讨丝裂霉素联合卡介苗(BCG)激活的淋巴细胞对膀胱肿瘤细胞株TBC-1的杀伤作用.②方法 丝裂霉素作用TBC-1约2小时后,观察在不同时间点热休克蛋白70(HSP70)的表达情况,加入经BCG刺激的淋巴细胞,检测时TBC-1的杀伤活性.③结果 丝裂霉素化疗TBC-1诱导HSP70的表达在0小时点表达最强,以细胞核为主,24小时表达最低,以细胞浆为主.丝裂霉素化疗TBC-1后0~8小时点加入BCG刺激的淋巴细胞杀伤作用最强.④结论 丝裂霉素联合BCG激活的淋巴细胞对膀胱肿瘤细胞株TBC-1有协同杀伤作用. Objective To study the killer effects of lymphocyte activited by mitomycin C with BCG on bladder carcinoma cell line TBC - 1. Methods The expression of HSP70 were measured at different time points after TBC - 1 was treated with mitomycin C for 2 hours, thereafter the killer effects of TBC - 1 were measured after adding lymphocytes activited by BCG. Reasults The expression of HSP70 in TBC - 1 treated with mitomycin C was the highest at 0 time point and mainly in nucleus, while which was the lowest at 24 hour time point and mainly in cell plasma. The killer effects of lymphocyte activited by BCG were the strongest after TBC - 1 was treated with mitomycin C at Oh - 8h. Conclusion There were the cooperative killer effects of lymphocyte activited by mitomycin C with BCG on bladder carcinoma cell line TBC - 1.
