摘要
目的通过测定游离血红蛋白(free hemoglobin,FHb)浓度以判别溶血程度。评价不同采血管及采血方式对血液溶血程度的影响。方法用新的2,4-二氯酚(2,4-DCP)法测定FHb浓度,比较用注射器采血和一次性采血针取血对血液溶血程度的影响,并比较枸橼酸钠、EDTA和肝素锂三种抗凝剂对溶血的影响。结果促凝管血清FHb浓度比肝素锂抗凝管[FHb=(25.2±5.6)mg/L]高。三种抗凝剂相比,枸橼酸钠(1:9)的血浆FHb浓度最低,FHb=(20.0±5.2)mg/L。一次性采血针FHb比注射器采血的FHb平均高25.8%。比较CG分离胶与CG促凝管,以进口BD管FHb最低。结论枸橼酸钠(1:9)抗凝剂管红细胞保存最好。促凝剂可致轻度溶血,BD管最好,分离胶管次之。各采血管3h后上升FHb浓度,24h后升高显著。
Objective In order to evaluate the different effects of blood collection tubes and drug additive, the authors determined the free haemoglobin (FHb) in plasm (serum). Methods Testing plasm (serum) free haemoglobin by new 2,4-dichlorophenol method to compare the two ways of syringe collect blood and vacuum blood collection blood between anticoagulated blood-collection tube and promote-coagulant tube. To compare vacuum blood collection ways in three anticoagulated blood-collection tubes. Resulte FHb concentration in promote-coagulant tube is higher than that in Lithium heparin anticoagulants tube [ FHb = (25.2 ± 5.6) mg/L ] . The FHb concentration of sodium citrate anticoagulant tube [ FHb = (20.0 ± 5.2)mg/L] is the lowest among three anticoagulated blood-collection tubes . FHb concentration in vacuum blood collection tube is 25.8% higher than syringe collect blood tube. FHb concentration of import BD tube is the lowest than gel vacuum tube and promote-coagulant tube. Conclusion The sodium citrate anticoagulant tube keeps erythrocyte best. The promote-coagulant may cause hemolysis very lightly. FHb concentrations in promote-coagulant tube is high than gel vacuum tube. BD tube is most lower. FHb concentration in all blood collection tubes rise after 3h, and increase after 24h obviously.
