2型糖尿病模型db/db小鼠胰岛B细胞超微结构观察 被引量:4

摘要 目的:观察2型糖尿病动物模型-C57BL/KsJ(db/db)小鼠的胰岛B细胞超微结构变化,探讨B细胞的病理改变在2型糖尿病发病中的作用。方法:分别选取3月、5月和8月龄尾静脉空腹血糖高于11.1mmol/L,且肥胖的db/db自发性糖尿病小鼠,每组5只作为糖尿病组;选取相应年龄段尾静脉空腹血糖低于6.0mmol/L,体重正常的db/+m表型正常小鼠,每组5只作为对照组。于相应年龄段取胰尾,透射电镜观察胰岛B细胞超微结构。结果:B细胞胞浆内的分泌颗粒数量明显减少,有的细胞甚至缺如,致密芯电子密度降低;线粒体数量减少,肿胀变形,粗面内质网不同程度扩张脱颗粒;细胞核不规则,染色质边集浓染;细胞间髓样小体增多,其严重程度随病情进展逐渐加重。结论:2型糖尿病动物模型db/db小鼠胰岛B细胞存在超微结构的病理改变,严重程度与病情的轻重有关,B细胞功能上的缺陷可能是2型糖尿病的一个重要因素。 Objective:To investigate the ultrastructural changes of the pancreatic islet B -cells of db/db diabetic mice. Methods:The test group selected 3,5 and 8 monthold db/db mice. The agematched db/+ m mice were as the control group. Every group included 5 mice. The pancreatic islet B - cells were observed under transmission electron microscope. Results: Insular B - cell structure of db/db mouse was becoming worse and worse as the disease developed. Numbers of secretory granule in B - cell cytoplasm were significantly decreasing or absent,and the electron - density of dense core in the secretory granule decreased as well. Mitochondria num- bers were decreasing and its volume was swollen. Rough endoplasmic reticulum was swollen, too. Cell nucleus was irregular. Chromatin was marginal and condensing. Myelin - bodies between insular cells were increasing. Conclusion:The study confirms that the pancreatic islet B - cells show ultrastructural changes in db/db diabetic mice,and uhrastructural change of B - cells is related to the degree of diabetes mellitus.
