番茄黑环病毒检测方法的建立 被引量:4

The research of detection methods for Tomato black ring nepovirus

摘要 通过汁液摩擦接种的方法接种几种鉴别寄主,筛选出检测番茄黑环病毒(TBRV)的一套鉴别寄主。同时建立了免疫学电镜检测方法。根据TBRV外壳蛋白基因的保守序列设计2对引物,应用RT-PCR均能从TBRV的6个分离物中扩增到与预期大小相同的DNA片段。利用抽提的质粒研究了RT-PCR方法检测TBRV的灵敏度。 The diagnostic species were selected using virous partile to inculate the indicator plants.Electron microscopy observation was established.RT-PCR amplification was established using two primers pair specific to the conserved sequence of TBRV coat protein gene produced a specific band with a expected size from the 6 isolates of TBRV.Sequence determination and analysis was showed that the determined sequence is partial TBRV coat protein gene.The Sensitivity of TBRV is determined using dilution of Plasmind of TBRV by RT-PCR.
