摘要
对同一时期三尖杉(Cephalotaxus fortunei)取材以及不同时期取材的灭菌效果进行对比试验,结果表明带芽茎段取材时期以3月中旬为最佳,此时萌发的新梢污染率较低、存活率较高,以75%酒精浸泡1 min,0.1%HgCl2灭菌12 min处理,存活率最高为63.3%,达到较好的灭菌效果。不同培养基与激素组合对三尖杉茎段腋芽诱导及增殖培养效果的试验,结果表明:6-BA质量浓度对带芽茎段启动速度、芽体诱导率影响极显著,改良MS+6-BA3.0 mg.L-1+IBA0.5 mg.L-1为三尖杉带芽茎段启动培养芽体分化的最佳培养基组合,芽体诱导率为82.8%;改良MS+6-BA2.0 mg.L-1+IBA0.2 mg.L-1为增殖培养最佳培养基组合,增殖系数为3.9。
The effect of sterilization of Cephalotaxus fortunei picking explants in the same period and the different period was measured. The results indicated that March was the best period for p/eking explants in which the pollution rate of the young stem was lower and the survival rate was higher. The hest sterilization procedure of stem with buds was surface disinfected with 75% ethanol for 1 min, followed by 0. 1% HgC12 for 12 rain, and the survival rate was 63.30/0. Different combinations of basal medium and hormone were studied on the induction and proliferation of the shoots. The concentration of 6-BA had greatly significant effect on the initial culture speed and the buds-induction rate of stems. The best initial medium for bnds-indueing of stems was modified MS + 6-BA 3.0 mg.L^-1 + IBA 0. 5 mg. L^-1 , and the inducement rate was 82. 8%. The best subculture medium for proliferation was modified MS +6-BA 2. 0 mg.L^-1 + IBA 0. 2 mg-L^-1 , and the proliferation coefficient was 3.9.
