摘要
目的:研究不同浓度集落刺激因子-1(CSF-1)、白细胞介素-1α(IL-1α)对大鼠牙囊细胞CSF-1受体基因表达的影响;明确CSF-1自分泌作用的抑制机制,为研究细胞因子在牙齿萌出和牙槽骨吸收中的作用奠定基础。方法:在培养并纯化的牙囊细胞培养液中分别加入不同浓度CSF-1、IL-1α,采用逆转录多聚酶链反应法(RT-PCR)检测这2种细胞因子对牙囊细胞CSF-1受体基因表达的影响。结果:高浓度CSF-1作用下,牙囊细胞CSF-1受体mRNA表达明显消失。不同浓度IL-1α作用后,CSF-1受体mRNA水平保持不变。结论:高浓度CSF-1抑制CSF-1受体基因的表达,不同浓度IL-1α对牙囊细胞CSF-1受体mRNA表达无影响。
AIM: To study the effects of colony stimulating factor( CSF - 1 ) and IL -1α on CSF - 1 receptor mRNA levels and to determine if the autocrine effect is inhibited through the CSF - 1 receptor . METHODS : Rat dental follicle cells were cultured in vitro. The effects of different concentrations of CSF - 1 and IL -1α on CSF - 1 receptor gene expression were detected by means of RT - PCR. RESULTS : High concentrations of CSF - 1 reduced the gene expression of the CSF - 1 receptor. IL -1α had no effects on CSF - 1 receptor mRNA levels. CONCLUSION: High concentrations of CSF - 1 inhibit the expression of CSF - 1 receptor. IL -1α has no effect on the expression of CSF - 1 receptor mRNA.
