摘要
目的探讨脊髓挤压伤后,BDNF在脊髓腹角和背角神经元的表达变化。方法各组大鼠脊髓做20μm厚冰冻横切片,免疫组化ABC法染片,灰度值测定;RT-PCR技术及凝胶成像系统分析BDNF mRNA的表达。结果BDNF主要分布于脊髓灰质神经元的胞浆中。在损伤后,腹角和背角的BDNF阳性神经元数均较正常组增高,3d组较正常组增高(P<0.01),7d和14d组较正常组则显著增多(P<0.01)。腹角和背角神经元的阳性灰度值均较较正常组降低,3d组的BDNF样免疫阳性灰度值较正常组降低(P<0.05),7d和14d组的BDNF较正常组降低更加显著(P<0.01),但7d组的BDNF阳性神经元数和灰度值有最显著的变化。正常组和损伤后各组的脊髓都表达BDNF mRNA,脊髓损伤后24小时组,BDNF mRNA表达达到高峰(P<0.01),以后它们的表达则逐渐下降,14d组与正常组已无显著差异(P>0.05)。结论在脊髓挤压伤后早期脊髓腹、背角神经元BDNF表达增强,提示内源性BDNF在脊髓损伤修复早期中发挥重要作用。
Objective To explore the expression changes of BDNF in the spinal cord ventral and dorsal horn neurons following crushed spinal cord injury(cSCI).Methods The expression of BDNF on the 20 μm transections of the spinal cord in rat was observed by immunohistochemical ABC method and quantitative grey density analysis.BDNF mRNA was analyzed by RT-PCR technique and gel imagine analysis system.Results The BDNF positive products were mainly distributed in cytoplams of neurons in the grey matter.The positive neurons counted in the ventral and dorsal horns for BDNF in 3d group following cSCI were higher than those in normal group(P〈0.05),in 7d and 14d groups following cSCI,the BDNF positive neurons counted in the ventral and dorsal horns were significantly higher than those in normal group(P〈0.01).Meanwhile,the quantitative grey densities of BDNF positive neuron in 3d group following cSCI were lower than those in normal group both in the ventral and dorsal horn(P〈0.05),in 7d and 14d groups following cSCI,they were significantly lower than those in normal group(P〈0.01),but the most markedly differences for the positive neurons and the quantitative grey densities of BDNF existed in 7d group.In addition,BDNF mRNA expression was observed both in the normal group and all other groups following cSCI,which reached its peak in 24h group(P〈0.01),and then the BDNF mRNA expression reduced gradually,no significant differences were detected between the 14d group and the normal group(P〉0.05).ConclusionBDNF expression was up-regulated in the spinal cord ventral and dorsal horn neurons following cSCI at early stage,which indicate that the increase of endogenous BDNF may play an important role in repair of nerve injury following cSCI.
