摘要
以蒂巴因为起始原料,经氧化、高压氢化、乙酰化、氰化、脱3-位甲氧基的甲基、水解、缩合、还原、成盐,合成了盐酸纳布啡,总收率达18.7%。改进的合成工艺原料易得、操作简便、成本低、中间产物质量得到控制、目标化合物质量符合药用要求,适宜于工业化大生产。
The synthesis of nalbuphine hydrochloride from thebaine as starting material was through following processes : oxidation, high-pressure hydrogenation, acetylation, removing methyl of 3-methoxy, hydrolysis, condensation,reduction and finial salification. The overall yield was 18.7%. This improved synthetic process had advantages of easy material, convenient operate and lower cost. Intermediate quality had been controlled effectively and target compound meeted medicinal quality standard. It was suitable for industrial production.
