摘要
应用流式细胞术检测大鼠腹腔感染经CF-11纤维素纯化的活弓形虫速殖子悬液后60 d内大鼠外周血CD3+、CD4+、CD8+T淋巴细胞的百分率,并应用酶联免疫吸附试验测定其外周血IFN-γ、TNF-α、IL-4等细胞因子的水平。结果显示:CD8+T细胞亚群水平在感染弓形虫后3 d、7 d、14 d和21 d时均显著低于正常水平(P<0.05),至感染后28 d恢复至正常;而CD4+T细胞亚群水平在整个感染过程中并无显著变化;CD4+/CD8+比值升高,以感染后14 d最为明显;大鼠外周血中IFN-γ和IL-4水平在感染后7 d开始升高(P<0.05),至感染后60 d时仍高于正常水平,TNF-α至感染后28 d时高于正常水平(P<0.05),至感染后60 d时仍高于正常水平。表明大鼠感染弓形虫对其免疫功能有一定程度的影响。
T lymphocytes play a key role in anti-infectious immunological response. In order to investigate the immunity of T lymphocytes of rat after being infected by Toxoplasma gondii, flow diagnosis of T cell subpopulations and ELISA of the levels of cytokines IFN-γ, TNF-α and IL-4 in the serum of rat infected with tachyzoites suspension of Toxoplasma gondii purified by cellulose CF-11 were performed. The amount of T cell subpopulation CD8 ^+ in the peripheral blood of infected rat was lower than (P 〈0. 05 ) that in non-infected rat on day 3, day 7, day 14 and day 21 of infection, and was increased to normal on day 28. Another subpopulation CD4 ^+ did not exhibit significant changes, however, the ratio of CD4^+ to CD8 ^+ rose to the highest on day 14 of infection. The elevation (P 〈0. 05 ) of the contents of cytokines IFN-γ, and IL-4 began on day 7 of infection and lasted to day 60. The content of IL-4 was found increased ( P 〈 0. 05 ) from day 28 to day 60 as well. Based on the results observed above, it was elucidated that the infection by Toxoplasma gondii could affect the immunological responses of rat.
