摘要 目的探讨PPARγ2基因Pro12ALa多态性与肥胖相关性。方法将研究对象按体质量指数(BMI)分为肥胖组(n=62)和非肥胖组(n=121)。再将所有对象分为糖尿病组(DM组,n=79)、对照组(NC组,n=104);DM组和NC组再分别根据BMI分为肥胖组与非肥胖组。应用聚合酶链反应-限制性内切酶片段长度多态性方法检测PPARγ2基因Pro12Ala多态性。比较肥胖与非肥胖组、DM组中的肥胖与非肥胖人群、NC组中的肥胖与非肥胖人群基因型频率及等位基因频率。结果肥胖组和非肥胖组基因型频率和等位基因频率无显著性差异(P>0.05)。进一步分组,DM组中肥胖组和非肥胖组基因型频率、等位基因频率之间无显著性差异(P>0.05),NC组中肥胖组和非肥胖组基因型频率无显著性差异(P>0.05),而等位基因频率有显著性差异(P<0.05)。结论PPARγ2基因Pro12Ala多态性与无T2DM家族史的正常人群肥胖有关,与糖尿病组中肥胖人群无关。 Objective To explore the correlation between Pro12Ala polymorphism of PPARγ2 in obesity. Methods One hundred and eigthy three patients were divided into two groups with the index of BMI. 62 people with obesity and 121 people without obesity. Further divided into two groups, the DM group consisted of 79 T2DM patients. The NC group consisted of 104 people as control. The method of PCR-RFLP was applied to research polymorphism of Pro12Ala of PPARγ2 in the DNA. Results There was no significant difference in the gene frequency and allelic gene frequency between the obesity group and non-obesity group(P〉0.05). In the DM subgroup, there was no significant difference in the gene frequency and allelic gene frequency(P〉0.05). In the NC subgroup, there was no significant difference in the gene frequency(P〉0.05), but there was significant difference in the allelic gene frequency between the obesity group and non-obesity group(P〈0.05). Conclusion Pro12Ala polymorphism in PPARγ2 might be related with obesity in Chinese without family history of T2DM.
