摘要
目的:探讨前列腺相关抗原-前列腺酸性磷酸酶(PAP)在非前列腺癌中的表达水平.方法:分别用RT-PCR方法和Western Blot方法检测多种腺癌细胞系中PAP mRNA和PAP蛋白的表达水平;用免疫组织化学方法检测多种腺癌肿瘤组织中的PAP蛋白的表达水平.结果:3种结肠癌细胞(colo201,colo205和colo320)、3种胃癌细胞(MKN-7,MKN-28和MKN-45)和7种乳腺癌细胞(OCUB-F,OCUB-M,R-27,MCF-7,CRL-1500,YMB-1-E和MDA-MB-231)表达PAP mRNA和PAP蛋白,结肠癌、胃癌和乳腺癌肿瘤组织中PAP呈阳性表达.在检测的5种肾细胞癌细胞和肾细胞癌组织中均未发现有PAP的表达.结论:PAP是结肠癌、胃癌和乳腺癌的新靶点.
AIM: To study the expression of prostatic acid phosphatase (PAP) , a prostate-related antigen, in non-prostate cancer cells or tissues. METHODS: A variety of adenocareinoma cell lines were examined for their PAP expression at the mRNA and protein levels by RT-PCR and Western Blot analysis, respectively. The PAP expression in cancer tissues was also examined by immunohistoehemieal staining. RESULTS: Three kinds of colon cancer cells (colo 201, colo 205 and colo 320 ) , 3 kinds of gastric cancer cells ( MKN-7, MKN-28 and MKN-45 ), and 7 kinds of breast cancer cell lines ( OCUB-F, OCUB-M, R- 27, MCF-7, CRL-1500, YMB-1-E and MDA-MB-231 ) as well as cancer tissues were found to be positive for PAP at both the mRNA and protein levels. But is was not found in 5 kinds of kidney cancer cells or tissues. CONCLUSION: PAP is a new target of non-prostate adenocarcinomas, including colon, breast and gastric cancers.
