摘要
[目的]探讨硝酸镧对大鼠生长及其肝脏中谷丙转氨酶(GPT),谷草转氨酶(GOT),碱性磷酸酶(AKP),γ-谷氨酰转肽酶(γ-GT)活性的影响。[方法]实验分为短期实验和长期实验,每期实验分为对照组和3个不同剂量组(0.1 mg/kg、2 mg/kg、20 mg/kg体重),灌喂硝酸镧,记录其体重和采食量变化。实验结束后,分别测定其肝匀浆中GPT、GOT、AKP、γ-GT活性。[结果]短期实验中,大鼠采食量与对照组相比变化不大,其增重率分别提高了11.33%、5.96%和2.14%,但均未达显著水平(P﹥0.05)。长期实验中,0.1 mg/kg和2 mg/kg组的大鼠,其日采食量与对照组相比变化不大,大鼠增重率分别提高了7.81%和9.94%,而20 mg/kg组大鼠采食量和增重率均降低。GPT酶活性在短期均表现出"低促-高抑"的效应;长期高浓度的镧降低GPT的活性。硝酸镧对GOT活性的影响主要表现在低剂量上,短期降低活性,长期有明显的增强作用。无论短期和长期,硝酸镧能提高AKP活性。γ-G活性则无显著变化。[结论]镧能影响大鼠的生长,影响肝脏GPT、GOT、AKP的活性,其结果与镧的剂量及灌喂长短有关,某些酶表现出低剂量促进,高剂量降低的hormesis效应。
[Objective]To investigate the effects of lanthanum nitrate on the growth and activities of several liver enzymes in rats.[Methods]40 SD rats were randomly divided into eight groups,with four groups for short-term(10 d)study,and the other four groups for long-term(30 d)study.The rats in each of the four groups were feeded with saline at 2 ml,lanthanum nitrate at the does of 0.1,2.0 and 20.0 mg/kg body weight,respectively.Daily food intake and weight gains were recorded.The activities of GPT,GOT,AKP and γ-GT were measured at the end of experiments.[Results]In short-term study,no significant difference in food intake were observed between experiment and control groups.Compared with control group,the weight gains of groups of lanthanum nitrate at the level of 0.1,2.0 and 20.0 mg/kg were improved by 11.33%,5.96% and 2.14%,respectively(P﹥0.05).In long-term study,there were also no changes in food intake between experiment and control groups,but the weight gains were improved by 7.18% and 9.94% in groups of lanthanum nitrate at the level of 0.1 and 2.0 mg/kg,while group of 20mg/kg had a little decrease in both food intake and weight gain.In short-term study,the activities of GPT in rat liver increased at low dose of lanthanum nitrate,and decreased at high dose of lanthanum nitrate,namely hormesis effect.In long-term study,lanthanum nitrate at high dose decreased its activity.Lanthanum at low dose could reduce the activity of GOT in short-term,but enhance it in long-term.However either short-term or long-term,lanthanum could enhance the activity of AKP.Meanwhile,γ-GT activity was not significantly affected by lanthanum nitrate.[Conclusion]Lanthanum nitrate can influence the growth of rats and the activities of GPT,GOT and AKP in rat liver.The effects depend upon the dose of lanthanum and the time of the experiment.Lanthanum has hormesis effect on activates of some liver enzymes.
