人β防御素3基因工程菌株的IPTG诱导条件优化 被引量:1

Optimization of Conditions for the Production of Human β Defensin 3 in E.coli Induced by IPTG

摘要 对人β防御素3基因工程菌株BL21-pET-hBD3的摇瓶生长和诱导条件进行了优化.对装液量、接种量和初始pH对菌种生长的影响进行了比较.结果表明,装液量对菌株生长影响最大.IPTG浓度、诱导时间和诱导温度对目的蛋白表达的试验结果显示,温度是一重要影响因素.进一步研究表明,菌株在37℃生长,在28-34℃以0.2 mmol.L-1IPTG诱导4 h对目的蛋白的表达有利,目的蛋白的含量最高可以达到总蛋白的29.8%. The cultural and inductive conditions for the flask-shaking of E. coli BL21-pET-hBD3 have been optimized. The effect of such factors as culture volume, inoculation volume, and initial pH value, on the growth was compared. The results showed that culture volume was found that culture volume was the most important influencing factor on the growth of strain. The influences of concentration of IPTG, time of induction and temperature on the yield of hBD3 were analyzed and studied in detail. The result indicated that the temperature was the most important factor. Further analysis showed that the more aduantageous expression level of hBD3, as high as 29.8% of total strain protein,could be gained when the strain was grown under 37℃and induced by 0.2 mmol·L^-1 IPTG under 28 - 34℃ for 4h.
