摘要
目的:检测c ide b蛋白在小鼠正常组织表达情况以及c ide b在小鼠胰腺中的确切定位,以期初步了解c ide b在胰腺组织中的生物学功能。方法:利用免疫组织化学———链霉素抗生物素-过氧化物酶(SP)法检测c ide b蛋白在小鼠正常组织中的表达分布;应用镜影切片染色和免疫荧光双标记染色及激光共聚焦扫描显微镜技术,观察c ide b和胰岛素在小鼠胰腺中的共定位情况。结果:c ide b蛋白除了在小鼠肝、小肠、肾等组织内表达外,在胰腺胰岛细胞中也有特异性表达;胰岛内c ibe b阳性表达细胞与胰岛素阳性细胞的分布有部分重叠的关系。结论:c ide b在小鼠胰岛β细胞特异表达,提示其可能与胰岛素的分泌、调节有关。
Objection: To detect the expression of cide b protein in mouse normal tissues and identify its exact localization in the mouse pancreas. Methods: The expression of cide b protein was detected in mouse normal tissues by immunohistochemistry (streptavidin-biotin-peroxidase, SP). The co-locational relationship between cide b and insulin in the mouse pancreas was studied by mirror sectioning technique, immunofluorescence double labeling staining and confocal laser-scanning microscope. Results: Not only did cide b protein express in the liver, small intestine, kidney, et al, but specifically in the islet cells of the pancreas. There was partial overlapping between the disposition of cide b and insulin positive cells. Conclusion :The specific expression of cide b protein mouse β islet cells indicated that cide b might be potentially associated with insulin secretion and regulation.
