摘要 目的采用HPLC-荧光检测方法测定大鼠脑组织中的单胺类递质及相关物质。方法脑组织匀浆用提取液(含0.1mol.L-1HClO4和0.2 mmol.L-1EDTA)提取后进样。色谱柱用Waters Nova-Pak C18(150 mm×3.9 mm,4μm);流动相为甲醇-缓冲液(14:86);流速1.0 ml.min-1;荧光检测波长λEX=280 nm,λEM=330 nm。结果7种单胺类神经递质及相关物质的标准曲线在一定浓度内线性关系良好,方法的加样回收完全,精密度好。结论所建方法简便、快速、准确,可用于脑组织中单胺类神经递质及相关物质的测定。 OBJECTIVE To develop a method for determining monoamine transmitters and related compounds in rat brain tissue by ion - pair HPLC with fluorometric detector. METHODS The brain tissues were homogenized in HClO4 (0.1 mol · L^- 1 ) and EDTA - 2Na(0. 2 mmol·L^-1). And the supematants were injected into the chromatographic system. The HPLC condition was Nova -pak C,s column( 150 mm ×3.9 mm, 4 μm), mobile phase:methanol - buffer( 14 : 86) ;flow rate:1. 0 ml·min^-1 ; fluorometric detector wavelength setting of excitation at 280 nm and emission at 330 nm. RESULTS The calibration curves of 7 monoamines and related compounds were linear within the limited concentration ranges. The methodology recovered fully, the within day RSD and between days RSD were low. CONCLUTION This method is simple, fast, sensitive and accurate and can be used to determine simultaneously the concentrations of monoamine transmitters and related compounds in rat brain tissue.
