平喘药对哮喘大鼠肺组织糖皮质激素受体的影响 被引量:3

Effects of Antiasthmatic Drugs on the Glucocorticoid Receptors in the Lung Tissue of Asthlmatic Rat

摘要 糖皮质激素是治疗哮喘炎症的最有效药物。糖皮质激素受体(GR)数量和功能的变化对其疗效可能有重要影响。我们观察了SD大鼠哮喘模型肺组织GR在哮喘发病过程中的动态变化及美喘清、舒喘灵、氟美松雾化吸入及腹腔注射氟美松对哮喘动物肺组织GR的影响,结果发现哮喘发作后第1天,GR数量明显增多;哮喘第3至14天又降至健康对照组水平,但亲和力有明显增加。氟美松吸入及腹腔注射均可使GR下降幅度增大,尤以腹腔注射更为显著。美喘清、舒喘灵治疗组GR下降较少。 In this study , we established SD rat asthma models with the method of active immune and provocation by ovalbumin. After consecutive daily challenges of asthma attack , we determined the Glucocorticoid receptors ( GR ) in the lung tissue of the models ( n = 44 ) with radio-ligand competitive combining assay and 3H-dexamethasone as the hot ligand . The results showed that the GR Bmax was 109. 19 ±23. 79 fmol/mg protein on the first day (n=8) ,which was significantly increased than that of the healthy control group ( 61. 67±18.1 3 , n=10. However , the GR Bmax of the models decreased dramatically since the 3rd day to 4 3.94±1 1.02fmol/mg protein , which was mild less than that of the controls ; but much less than the lst day. The GR Kd value of the models decreased significantly since the 3rd day compared with the controls. After the first asthma challenge , we treated the rat asthma models with oral procaterol , oral salbutamol , inhaled dexamethason ( Ih . D ) and intraperetoneal dexamethasonge ( Ip. D ) daily. On the 7th day , we measured the GR Bmax (fmol/mg protein) were 70. 61 ±14. 86 , 68 , 1 2 ±12. 17 , 55. 27 ±20. 83 and 36. 58 ± 18. 84 , respectively. The Kd value had no significant changes. Indicating that the Beta-agonists might protect the GR from down regulation. The Ih . D had less effect on GR level than that of Ip. D. It is known that corticosteroids can down regulate their receptors. In our assay , we did not find low-affinity GR both in control and asthma model groups.
