摘要
目的探讨PI-3K/Akt信号通路在大鼠心脏缺血-再灌注损伤中的作用。方法选取雌性Wistar大鼠建立大鼠心脏缺血/再灌注损伤(I/R)模型,分为正常对照组,I/R组和I/R+LY294002组。I/R+LY294002组大鼠再灌注前腹腔注射LY294002 6 mg/kg。检测各组Akt的表达、大鼠心肌梗死面积、心功能和心肌细胞凋亡的变化。结果在大鼠心脏I/R中PI-3K/Akt信号通路被激活,应用LY294002后Akt表达减少,心肌梗死面积增加(30.47%±5.88%vs 45.97%±4.22%,P〈0.05),心肌细胞凋亡增加(20%±4.1%vs 35%±5.3%,P〈0.05),心功能恶化。结论在大鼠心脏I/R过程中,PI-3K/Akt信号通路被激活并发挥心脏保护作用。
Objective To investigate the effect of PI-3K/Akt signal pathway in rat heart ischemia and reperfusion (I/R) injury. Method Female Wistar rats were used to construct the heart ischemia and reperfusion (I/R) model. Groups included normal control group, I/R group and I/R plus LY294002 group, Western Blot was used to detect the Akt expression in the heart, The infarct size, heart function and apoptosis of the cardiomyocytes were detected in different groups of the rats. Result PI-3 K/Akt signal pathway was activated in I/R injury heart of the rats, After administrated of LY294002, the level of Akt expression was significantly decreased. Simultaneously, the injuries of the heart became severer than before. Condusion PI-3K/Akt signal pathway was activated in I/R injury in rat heart, which could protect the heart from I/R injury.
