循环DNA定量分析在乳腺癌诊断中的应用 被引量:2

摘要 目的:检测乳腺癌患者循环DNA含量,探讨其在肿瘤诊断中的价值。方法:收集61例乳腺癌患者、33例乳腺良性病变患者和20例健康志愿者的血浆样本,以SYBRgreenI荧光染色法抽提血浆循环DNA,行DNA定量分析。结果:乳腺癌患者、乳腺良性病变患者、健康志愿者循环DNA浓度分别为65.46士15.31ng/ml、19.29士9.54ng/ml、13.56士7.39ng/ml;乳腺癌患者显著高于乳腺良性病变患者和健康志愿者,差异有显著性意义(P<0.05)。结论:血浆肿瘤循环DNA定量分析在乳腺癌诊断中有较高的应用价值。 Objective: To quantify the circulating DNA in plasma from patients with breast cancer and to explore its value in tumor diagnosis.Method: Blood samples were collected from 61 breast cancer patients, and from 33 patients with non-neoplastic breast diseases, and 20 healthy volunteers.The circulating DNA was extracted and analyzed by fluorenscent (SYBR green I) staining. Results: The circulating DNA concentration in the breast cancer patients was 65.46±15.31 ng/ml, while that in non-neoplastic breast diseases patients was 19.29士9.54 ng/ml,in the normal control was 13.56士7.39 ng/ml.Significant difference (P<0.05) in the circulating DNA levels was found between breast cancer patients and the other two groups.Conclusion: Quantitative analysis of circulating DNA level in plasma has an important diagnosis value to the breast cancer patients.
