摘要
对1株苹果轮纹病细菌No.1菌株的发酵条件进行了探索,确定了其最佳培养基组成及培养条件。单因素发酵试验和正交试验结果表明,在含酵母浸膏5g、蔗糖20g、蛋白胨10g的1000mL液体培养基中,菌体生长量最大;细菌生长所需的最佳碳源为淀粉,最佳氮源为酵母浸膏和蛋白胨;当温度为28℃,pH7.0,在250mL三角瓶中装40mL液体培养基,接种量体积分数为5%,培养时间96h时,菌体生长量最大;在酸性条件下(pH4),细菌几乎不生长。
The fermentable conditions of antagonistic bacteria No.1 strain were studied. The composition of fermentation medium and cultural conditions were confirmed. The single factor and orthogonal experiment indicated that the optimal cultural medium was a liquid medium (yeast extract 5 g, sucrose 20 g, protein peptone 10 g). The most suitable carbon source was starch. The most suitable nitrogen sources were peptone and yeast extract; the optimal culture conditions were 28 ℃, pH 7.0, 40 mL/250 mL flask of culture volume, 96 h culture time and 5% inoculated volume. At pH 4 the antagonistic bacteria almost not grew.
