摘要
目的:探讨HSPC300蛋白与原发性结肠癌发生与发展的关系,及其异常表达的细胞生物学意义。方法:用免疫组织化学方法分析HSPC300蛋白在正常、腺瘤及结肠腺癌中的表达;通过体外细胞迁移系统观察异常表达的HSPC300对结肠癌细胞迁移能力的影响。结果:HSPC300在正常结肠组织中的表达阳性率为25%(5/20),在结肠腺瘤组织中为15%(3/20),结肠癌组织中为68%(15/22)。HSPC300在结肠正常组织及腺瘤组织中的表达频率显著低于腺癌(P=0.0002)。体外迁移实验表明,下调结肠癌细胞中HSPC300的表达可降低其迁移能力。结论:HSPC300在结肠癌的发生、发展过程中是一个晚期分子事件,其高表达有助于结肠癌细胞获得转移潜能。
Objective: To study the expression distribution of HSPC300 in colonic normal, adenoma and carcinoma tissues, and migration of carcinoma cell in vitro. Methods: By tisuue immunochemistry, the expression of HSPC300 in colonic normal, adenoma and carcinoma tissues was detected. By the system of cell migration in vitro, the effect of abnor- mally expressed HSPC300 to the migration of colonal cancer cell was also observed. Results: The results showed that ex- pression of HSPC300 elevated significantly in colon carcinoma tissue, compared with those in normal and adenoma tissue. Down-regulated expression of HSPC300 by siRNA would inhibit the migration of colonal cancer cell in vitro. Conclusion: It indicated that the up-regulated expression of HSPC300 may contribute to the progress of colonal cancer.
